The ADC binds Nectin-4 on tumor cells, is internalized, and releases MMAE, which binds beta-tubulin to disrupt microtubules and induce apoptosis. Beta-tubulin is the intracellular payload target, not the targeting antigen; expression of beta-tubulin alone does not result in killing without Nectin-4–mediated delivery.
