The ADC targets B7-H3 on tumor cells, is internalized, and releases a topoisomerase inhibitor payload that kills those B7-H3–positive cells by blocking DNA replication and inducing apoptosis. DNA topoisomerase is the intracellular payload target, not the cell-selection target; its expression alone does not lead to killing.
