Sacituzumab govitecan binds Trop-2 (not DNA topoisomerase I), is internalized, and releases SN-38, which inhibits topoisomerase I to cause DNA breaks, cell-cycle arrest, and apoptosis; the membrane-permeable payload can also produce bystander killing.
