The ADC binds HER2 on tumor cells, is internalized, and releases MMAE, which then binds beta-tubulin (vinca site) to disrupt microtubules and trigger G2/M arrest and apoptosis. Beta-tubulin expression alone does not lead to targeting or killing without HER2-mediated delivery.
