Belantamab mafodotin targets BCMA on plasma cells; after internalization, the MMAF payload binds tubulin (vinca domain) to block microtubule polymerization, causing G2/M arrest and apoptosis, with additional ADCC/ADCP. Cells are not killed based on tubulin expression alone.
