Brentuximab vedotin binds CD30 on the cell surface, is internalized, and releases MMAE, which then binds beta‑tubulin (vinca domain) to block microtubule polymerization, causing G2/M arrest and apoptosis. Tubulin expression alone does not confer killing; CD30-mediated delivery is required.
